TBF BlockerTM Tissue Background Fluorescence Blocker

(IC-20020)

Description

Many tissues produce endogenous spontaneous fluorescence that can pass through various wavelength filters, significantly interfering with antibody labeled fluorescence observation and even leading to immunohistochemical staining failure. TBF BlockerTM can capture electrons emitted by spontaneous fluorescence light source molecules through collision, preventing the electrons from returning from the excited state to the ground state and releasing energy, thereby quenching spontaneous fluorescence. By optimizing the incubation time, background fluorescence can be eliminated to the maximum extent without significantly affecting the fluorescence of antibody labeling.TBF BlockerTM is suitable for the spontaneous fluorescence elimination of immune fluorescence staining in various tissues and cells. Especially suitable for spontaneous fluorescence quenching of neural tissues.

Storage

2-8¡æ for 12months.

Usage

1. Remove PBS or corresponding washing buffer and briefly rinse the cells in tissue slices or cell culture plates with dd H2O.

2. Add an appropriate amount of TBF BlockerTM to cover cells in tissue slices or bottles, and incubate at room temperature for 10-90 minutes.

Note:This step can optimize the incubation time to achieve the best quenching effect without significantly reducing the fluorescence intensity of the antibody.

3. Remove the TBF BlockerTM  and rinse briefly with dd H2O.

4. Remove dd H2O and cover tissue slices or cells located in cell culture plates with PBS.

5. Drop a drop of Mounting Medium,Antifading(IC-20016) on the tissue slice, cover the cover glass, let the slice contact the sealing liquid, and try to avoid bubbles. Mounting Medium,Antifading can prevent antibody labeled fluorescence decay.

6. The sample can then be observed with a ( Confocal ) fluorescence microscope .

Please Note Information
1. The background fluorescence of different types of tissues has different characteristics, and the fluorescence quenching effect using TBF BlockerTM may vary.

2. The quenching of background fluorescence by TBF BlockerTM  will to some extent reduce the fluorescence intensity of the antibody.

3. The test results of this product show that it has a better effect on eliminating background fluorescence of brain and spinal cord nerve tissue.

4.TBF BlockerTM  must be used after completing immunofluorescence histochemical staining, otherwise it will severely reduce antibody fluorescence.

Important Statement
For precious samples, it is necessary to optimize them on ordinary samples before using this reagent to avoid irreparable losses of precious samples. Optimization principle: The background fluorescence decreases with the prolongation of the incubation time of the product, while the antibody fluorescence intensity increases with the shortening of the incubation time of the product.
