This product is a cell membrane labeling kit based on the lipophilic fluorescent dye PKH26. PKH26 contains long aliphatic chains that stably insert into the lipid bilayer of the cell membrane, providing stable, uniform, and non-specific fluorescent labeling of living cells.

Protocol (For Reference Only)
Important: Staining must be performed under serum-free conditions.
1. Cell Preparation: Prepare a single-cell suspension, wash cells twice with serum-free medium or PBS, and resuspend pellet in Diluent C (provided) at 1x10^6 to 1x10^7 cells/mL.
2. Dye Working Solution (Prepare Fresh): Dilute PKH26 stock solution in Diluent C to achieve a final concentration of 2-4 µM during staining and mix gently.
3. Staining: Rapidly mix equal volumes of cell suspension and 2X Dye Working Solution, incubate at room temperature (25¡ãC) for 2-5 minutes, and gently invert periodically. Note: Avoid prolonged incubation.
4. Stop Reaction: Add an equal volume of Fetal Bovine Serum (FBS) or complete medium containing 10% FBS, incubate for 1 minute at RT, then centrifuge and wash cells 2-3 times with complete medium until supernatant is clear.
5. Analysis: Resuspend cells in complete medium for flow cytometry, microscopy, culture, or injection.
