Storage and Stability

Protocol (For Reference Only)

Important: Use aseptic technique when handling medium to prevent contamination. Warm medium to 37¡ãC before use for optimal cell growth.

1. Medium Preparation: Warm Mouse Cardiac Fibroblast Immortalized Cell Culture Medium to 37¡ãC in a water bath or incubator for 15-30 minutes before use. Do not add additional serum or supplements as this medium is a complete formulation.

2. Cell Thawing: Thaw frozen cells rapidly in a 37¡ãC water bath. Transfer the cell suspension to a centrifuge tube containing 5-10 mL of pre-warmed medium. Centrifuge at 200 x g for 5 minutes, discard supernatant, resuspend in fresh pre-warmed medium, and seed into appropriate culture vessels.

3. Routine Culture: Incubate cells at 37¡ãC in a humidified 5% CO₂ incubator. Replace medium every 2-3 days or when the pH indicator (phenol red) shifts from pink-red to yellow-orange.

4. Subculturing: When cells reach 80-90% confluency, aspirate medium, wash with PBS, and detach cells using 0.25% trypsin-EDTA at 37¡ãC for 2-3 minutes. Neutralize trypsin with fresh complete medium, centrifuge at 200 x g for 5 minutes, resuspend in fresh medium, and seed at an appropriate split ratio (typically 1:3 to 1:5).

5. Cryopreservation: For cell banking, harvest cells at 80-90% confluency, centrifuge at 200 x g for 5 minutes, resuspend in freezing medium (complete medium supplemented with 5-10% DMSO), aliquot into cryovials, and freeze at -80¡ãC overnight before transferring to liquid nitrogen for long-term storage.
