Product Introduction

This product is a polyvinylidene fluoride (PVDF) blotting membrane with a pore size of 0.45 µm, designed for protein transfer and immunodetection in Western blotting applications. PVDF membranes offer high protein binding capacity, mechanical strength, and chemical resistance, making them the preferred choice for Western blotting, protein sequencing, and other protein analysis techniques. The 0.45 µm pore size is suitable for proteins with molecular weights above 20 kDa, providing an optimal balance of binding capacity and low background. The membrane is supplied in convenient roll format, allowing users to cut membranes to the desired size. PVDF membranes must be pre-wetted with methanol before use to ensure proper hydration and protein binding.

Storage and Stability

Storage Conditions: Store at room temperature in a dry environment, protected from dust and direct sunlight.

Protocol (For Reference Only)

Important: PVDF membrane must be pre-wetted with 100% methanol before use. Do not allow the membrane to dry out during the transfer and detection procedure.

1. Membrane Activation: Immerse the PVDF membrane in 100% methanol for 15-30 seconds until the membrane becomes uniformly translucent. Transfer the membrane to deionized water and rinse briefly, then equilibrate in transfer buffer for at least 5 minutes.

2. Transfer Assembly: Assemble the transfer sandwich according to standard Western blot protocols, placing the activated PVDF membrane against the gel.

3. Protein Transfer: Perform electrophoretic transfer according to standard protocols using appropriate transfer buffer (e.g., Towbin buffer with or without methanol).

4. Post-Transfer Processing: After transfer, proceed directly to blocking and immunodetection. If desired, Ponceau S staining may be performed to assess transfer efficiency.

5. Stripping and Reprobing: PVDF membranes can be stripped of antibodies using appropriate stripping buffers and reprobed multiple times.

 

Precautions

1. Always pre-wet PVDF membrane with methanol before use; failure to do so will result in poor protein binding.

2. Do not allow the membrane to dry out during transfer and detection steps; if drying occurs, re-wet with methanol and re-equilibrate.

3. Wear gloves and use clean forceps to handle the membrane to avoid contamination.

4. Cut membranes to size before activation for clean edges and optimal results.
