Product Introduction

This product is a rapid blocking buffer (ready-to-use, wash-free), a protein-free blocking solution specifically designed for Western Blot. Traditional blocking solutions commonly employ animal-derived proteins such as non-fat dry milk, bovine serum albumin, or casein. While capable of saturating non-specific binding sites on the membrane, these protein-based blockers may contain phosphatase activity or exogenous immunoglobulins, potentially causing high background or non-specific bands in the detection of post-translational modifications such as phosphorylated or acetylated proteins. This product utilizes a fully synthetic, protein-free formulation without animal-derived components, capable of efficiently blocking the exposed surfaces of PVDF and NC membranes. The blocking process requires only 10 minutes, and primary antibody incubation can proceed directly after blocking without a washing step, simplifying the workflow and reducing experimental time. It is suitable for conventional antibodies, phospho-specific antibodies, and biotin-labeled antibodies. This product is formulated in TBST containing 0.1% TWEEN 20.

Protocol (For Reference Only)

1. Post-Transfer Wash: After completing protein transfer, wash the membrane with Western washing buffer for 1-2 minutes.

2. Blocking: Pour an appropriate volume of Rapid Blocking Buffer into a suitable container to ensure complete coverage of the membrane. For a standard Western blot, approximately 10 mL of buffer is recommended for a membrane of approximately 6.6 ¡Á 8.5 cm. Using flat-tipped forceps, grasp the membrane by a corner and place it into the blocking buffer, ensuring the membrane is completely immersed. Incubate on a horizontal shaker at room temperature for approximately 10 minutes.

3. Primary Antibody Incubation: After blocking, discard the blocking buffer and add primary antibody working solution directly without washing.

Precautions

1. Blocking Time: The standard blocking time is 10 minutes. For antibodies that produce high background, the blocking time may be extended to 30-60 minutes, or blocking can be performed overnight at 4¡ãC.

2. Reuse: Single use of the blocking buffer is recommended. For antibodies with high signal-to-noise ratios, such as certain housekeeping protein antibodies, the buffer may be reused 2-3 times. Do not mix used buffer with unused buffer.

3. Membrane Handling: Use flat-tipped forceps to handle PVDF and NC membranes, gently grasping only the corners. Avoid creating scratches, creases, or indentations on the membrane surface.

4. Membrane Condition: Once PVDF membranes have been wetted and activated, they must remain moist at all times. Drying may result in high background that is difficult to resolve.

5. Application Scope: This product is for membrane blocking only and should not be used for primary or secondary antibody dilution.

FAQ (Simplified)

Q1: Is 10 minutes of blocking really sufficient?

A1: Testing with various antibodies has demonstrated that a 10-minute block typically achieves results comparable to traditional BSA blocking for 1 hour. For individual antibodies with high background, the blocking time may be extended to 30-60 minutes.

Q2: Why is no wash step needed after blocking?

A2: This product features a protein-free formulation. Residual blocking buffer does not cross-react with primary antibodies, so the blocking buffer can be discarded and primary antibody applied directly without an intermediate wash step.

Q3: Can the buffer be reused?

A3: Single use is recommended. For antibodies with high signal-to-noise ratios such as housekeeping protein antibodies, the buffer may be reused 2-3 times. However, do not mix recovered buffer with fresh buffer, and performance may slightly decrease with reuse.

Q4: Can this product be used for primary or secondary antibody dilution?

A4: No. This product is intended for the blocking step only. Primary and secondary antibodies should be diluted according to the diluent recommendations in their respective instructions.
