Product Introduction

Macrophages are highly involved in maintaining physiological homeostasis and orchestrating pathological tissue responses. Macrophages exhibit extensive morphological and functional heterogeneity across physiological and pathological contexts including muscle motor control, cellular and vascular aging, obesity-related epigenetic imprinting, hematopoietic clonal quality control, infection and inflammation, tumor initiation and progression, metabolic and chronic diseases, and epigenetic reprogramming. The morphology and function of macrophages at early, intermediate, and late stages reflect a complex, dynamic, and continuous biological response process regulated in time and space by a series of immunological network microenvironments.

Clodronate liposomes are a widely used reagent for macrophage depletion. By encapsulating clodronate within liposomes, the phagocytic properties of macrophages are exploited to enable uptake of clodronate liposomes. After phagocytosis, clodronate is released within the macrophage lysosome through the action of lysosomal phosphatases. Accumulation of clodronate beyond a threshold specifically induces macrophage apoptosis, thereby depleting macrophages in tissues.

Control liposomes have the same properties as clodronate liposomes except that they do not encapsulate the active pharmaceutical ingredient clodronate. After being phagocytosed by macrophages, control liposomes do not induce macrophage apoptosis or depletion and serve primarily as a negative experimental control.

The Cell-Del® Clodronate Liposome Macrophage Depletion Set contains one vial of Clodronate Liposomes and one vial of Control Liposomes of equal volume. Administration routes are flexible and include intravenous, intraperitoneal, subcutaneous, intranasal, and intratracheal routes. In vivo injection doses should be determined based on animal body weight, injection frequency, administration route, and experimental objectives.

Protocol (For Reference Only)

Important: This product is a suspension. Invert several times before use to ensure uniform mixing. Do not aliquot. Do not freeze. Avoid contact with organic reagents and surface detergents.

1. Preparation: Remove the vial from 4-8¡ãC storage and invert several times to resuspend the liposomes uniformly.

2. Administration Route Selection: Select the appropriate administration route based on experimental objectives (intravenous, intraperitoneal, subcutaneous, intranasal, or intratracheal).

3. Dose Determination: Determine the injection dose based on animal body weight, injection frequency, administration route, and experimental objectives. Consult published literature for reference doses.

4. Injection: Draw the suspension using a sterile syringe under aseptic conditions. Inject according to the selected route.

5. In Vitro Experiments: For in vitro experiments, the liposome suspension may be diluted with sterile PBS.
