L-Lactate Dehydrogenase, E. coli, IC-03997865

Product Introduction

L-Lactate dehydrogenase (L-LDH) from Escherichia coli is a redox enzyme that catalyzes the reduction of pyruvate to L-lactate by NADH in vivo with absolute enantiospecificity. It is widely used in biochemical research, chiral synthesis, and enzymatic assays for lactate detection and quantification.

Product Features

1. High Enantiospecificity: Catalyzes the reduction of pyruvate to L-lactate with absolute enantiospecificity.

2. High Specific Activity: ¡Ý30 U/mg solid.

3. Good Solubility: Solubility in H₂O ¡Ý100 mg/mL.

4. Well-Characterized Properties: Defined molecular weight, isoelectric point, optimal pH and temperature, and stability profiles.

5. Ready-to-Use: Soluble in pure water for convenient preparation.

Protocol (For Reference Only)

Important: Dissolve in pure water. Avoid repeated freeze-thaw cycles.

1. Reconstitution: Dissolve L-Lactate dehydrogenase in pure water to the desired concentration. Mix gently; do not vortex vigorously.

2. Enzymatic Assay: Prepare reaction buffer at the optimal pH (6.5) containing NADH and pyruvate. Initiate the reaction by adding L-LDH. Monitor NADH oxidation spectrophotometrically at 340 nm.

3. Reaction Conditions: Optimal temperature is 45¡ãC and optimal pH is 6.5. For standard assays, incubate at 37¡ãC or as appropriate for the specific application.

4. Storage of Working Solution: Aliquot the reconstituted enzyme into single-use volumes and store at -20¡ãC. Avoid repeated freeze-thaw cycles.

Precautions

1. Avoid contact with inhibitors including Co²⁺, Cu²⁺, Fe³⁺, Ni²⁺, Zn²⁺, NEM, SDS, and Proclin.

2. Dissolve in pure water; avoid buffers containing inhibitory metal ions.

3. Aliquot into single-use volumes to avoid repeated freeze-thaw cycles.

4. For research use only. Not for use in diagnostic or therapeutic procedures.

FAQ (Simplified)
