Organoid Cryopreservation Medium, SF1800-06
Product Introduction
Organoid Cryopreservation Medium is suitable for the long-term low-temperature storage of organoids and cell lines derived from various mammalian species, such as human and mouse.
Organoid (Cell) Cryopreservation
(1) Add pre-chilled (2-8¡ãC) Organoid Cryopreservation Medium to the recovered organoids (cells), mix by pipetting, and immediately transfer to a cryovial (the cell suspension volume in the cryovial should be greater than 0.5 mL). Note: Each 1 mL of cryopreservation medium is recommended for 1¡Á10³ to 1¡Á10⁷ cells or an equivalent amount of organoids.
(2) Place the cryovial into a cell freezing container (the container must be pre-equilibrated to room temperature or 4¡ãC), then immediately place the container in a -80¡ãC ultra-low temperature freezer. Alternatively, perform manual gradient cooling: stand at 4¡ãC for 10 minutes, store at -20¡ãC for 1 hour, and store at -80¡ãC overnight.
(3) The next day or after 12 hours, transfer the cryovial under low-temperature conditions (not higher than -70¡ãC; dry ice or the original freezing container is recommended) to liquid nitrogen for long-term storage.
Organoid (Cell) Recovery
(1) Add 10 mL of Organoid Passaging Buffer (or PBS) to a 15 mL centrifuge tube.
(2) Remove the frozen organoids/cells from the liquid nitrogen tank and quickly place in a 37¡ãC water bath to thaw.
(3) During thawing, gently shake the cryovial to ensure complete thawing of the cryopreservation medium within 1-2 minutes.
(4) Quickly transfer the thawed organoids to the 15 mL centrifuge tube, gently pipette 6-8 times, centrifuge at 300 x g for 5 minutes, then remove the supernatant and collect the organoid/cell pellet. Add an appropriate volume of Organoid Passaging Buffer (or PBS) to resuspend, transfer to a 1.5 mL centrifuge tube, and centrifuge at 300 x g for 5 minutes.
(5) Resuspend in 20-25 times the centrifuged organoid volume of Matrigel, seed 25 µL of Matrigel per well in a 24-well cell culture plate, place in the incubator for 10-15 minutes to solidify, and add 500-750 µL of organoid culture medium.

Protocol (For Reference Only)
Important: Pre-chill the cryopreservation medium to 2-8¡ãC before use. Ensure the freezing container is pre-equilibrated to room temperature or 4¡ãC before use.
1. Cryopreservation:
1.1 Add pre-chilled (2-8¡ãC) Organoid Cryopreservation Medium to the recovered organoids (cells), mix by pipetting, and immediately transfer to a cryovial (cell suspension volume >0.5 mL). Each 1 mL of cryopreservation medium is recommended for 1¡Á10³ to 1¡Á10⁷ cells or an equivalent amount of organoids.
1.2 Place the cryovial into a pre-equilibrated cell freezing container, then immediately place in a -80¡ãC freezer. Alternatively, perform manual gradient cooling: 4¡ãC for 10 minutes, -20¡ãC for 1 hour, -80¡ãC overnight.
1.3 The next day or after 12 hours, transfer the cryovial under low-temperature conditions (not higher than -70¡ãC) to liquid nitrogen for long-term storage.
2. Recovery:
2.1 Add 10 mL of Organoid Passaging Buffer (or PBS) to a 15 mL centrifuge tube.
2.2 Remove the frozen organoids/cells from liquid nitrogen and quickly place in a 37¡ãC water bath to thaw.
2.3 Gently shake the cryovial during thawing to ensure complete thawing within 1-2 minutes.
2.4 Quickly transfer the thawed organoids to the 15 mL centrifuge tube, gently pipette 6-8 times, centrifuge at 300 x g for 5 minutes, remove supernatant, and collect the pellet. Resuspend in an appropriate volume of Organoid Passaging Buffer (or PBS), transfer to a 1.5 mL centrifuge tube, and centrifuge at 300 x g for 5 minutes.
2.5 Resuspend in 20-25 times the organoid pellet volume of Matrigel, seed 25 µL per well in a 24-well plate, allow to solidify in the incubator for 10-15 minutes, and add 500-750 µL of organoid culture medium.
Precautions
1. Pre-chill the cryopreservation medium to 2-8¡ãC before use.
2. Ensure the freezing container is pre-equilibrated to room temperature or 4¡ãC before use.
3. Transfer cryovials to liquid nitrogen under low-temperature conditions (not higher than -70¡ãC) to maintain viability.
4. Avoid repeated freeze-thaw cycles of the cryopreservation medium.
5. For research use only. Not for use in diagnostic or therapeutic procedures.
FAQ (Simplified)
Q1: What species are suitable for this cryopreservation medium?
A1: This medium is suitable for organoids and cell lines derived from various mammalian species, including human and mouse.
Q2: What is the recommended cell density for cryopreservation?
A2: Each 1 mL of cryopreservation medium is recommended for 1¡Á10³ to 1¡Á10⁷ cells or an equivalent amount of organoids.
Q3: Can I use manual gradient cooling instead of a freezing container?
A3: Yes. Manual gradient cooling (4¡ãC for 10 minutes, -20¡ãC for 1 hour, -80¡ãC overnight) may be used as an alternative to a controlled-rate freezing container.
Q4: How should I store the cryopreservation medium?
A4: Store at 2-8¡ãC or -20¡ãC according to product label. Pre-chill to 2-8¡ãC before use. Avoid repeated freeze-thaw cycles.
