Product Introduction
Defined Keratinocyte-SFM is a serum-free medium suitable for the isolation and expansion of human keratinocytes without the need for bovine pituitary extract (BPE) or fibroblast feeder layers. The growth-promoting activity of BPE is replaced by growth promoters added to the medium, resulting in more consistent performance. These growth promoters also maintain cell morphology, biomarkers, and growth characteristics. This medium suppresses fibroblast proliferation and has a very low calcium concentration (<0.1 mM), making it suitable for the isolation and culture of high-density undifferentiated keratinocytes.

Protocol (For Reference Only)
Important: Use aseptic technique throughout. Warm complete medium to 37¡ãC before use.
1. Complete Medium Preparation: If components are provided separately, thaw the supplements according to the specified storage conditions, then add to the basal medium under aseptic conditions. Mix gently by swirling.
2. Medium Pre-warming: Warm the required volume of complete medium to 37¡ãC for 15-30 minutes before use.
3. Cell Culture: Seed keratinocytes in appropriate culture vessels pre-coated with extracellular matrix if required. Incubate at 37¡ãC, 5% CO₂.
4. Medium Change: Replace medium every 2-3 days or when pH indicator changes color.
5. Subculturing: When cells reach the desired confluency, detach using trypsin-EDTA and passage according to standard protocols for keratinocytes.
