Goat Anti-Mouse IgG1-AF594, SA-59401-1
Product Introduction
This product is a goat polyclonal secondary antibody conjugated to AcelFluor 594 (AF594), specifically recognizing the heavy chain of mouse IgG1. Secondary antibodies are essential reagents in immunofluorescence (IF), immunohistochemistry (IH), flow cytometry (FC), ELISA (E), and other immunodetection techniques, providing specific detection of primary antibodies bound to target antigens. AF594 is a bright, photostable orange-red fluorescent dye with excitation/emission maxima of approximately 590/617 nm, compatible with standard TRITC/Texas Red filter sets and 561 nm laser-equipped instruments. The antibody is isolated from antisera by immunoaffinity chromatography using antigens coupled to agarose beads, ensuring high specificity and minimal cross-reactivity. By immunoelectrophoresis and ELISA, this antibody reacts specifically with mouse IgG1 heavy chain.

Protocol (For Reference Only)
Important: Protect fluorescent conjugate from light. Centrifuge the vial briefly before opening. Optimal working dilution should be determined empirically for each application.
1. Immunofluorescence (IF): Fix and permeabilize samples according to standard protocols. Block with appropriate blocking buffer. Incubate with mouse IgG1 primary antibody. Wash and incubate with AF594-conjugated secondary antibody at 1:100 to 1:1,000 dilution for 1 hour at room temperature in the dark. Wash and mount with anti-fade mounting medium.
2. Immunohistochemistry (IH): Process tissue sections according to standard protocols. Following mouse IgG1 primary antibody incubation and washing, apply AF594-conjugated secondary antibody at 1:100 to 1:1,000 dilution for 30-60 minutes at room temperature in the dark. Wash and mount.
3. Flow Cytometry (FC): Following cell staining with mouse IgG1 primary antibody, wash cells and incubate with AF594-conjugated secondary antibody at 1:1,000 to 1:4,000 dilution for 30 minutes at 4¡ãC in the dark. Wash and analyze using a 561 nm laser with appropriate emission filter.
4. ELISA (E): Dilute the AF594-conjugated secondary antibody according to assay requirements and incubate following primary antibody binding. Detect according to the specific ELISA format.
Precautions
1. Protect from light to prevent photobleaching of the AF594 fluorophore.
2. Avoid freeze/thaw cycles; aliquot upon delivery and store at -20¡ãC.
3. The recommended dilutions are guidelines; optimal dilution must be determined empirically for each application.
4. Sodium azide is included as a preservative; handle with appropriate precautions.
5. For research use only. Not for use in diagnostic or therapeutic procedures.
