Goat Anti-Mouse IgG2a-AF647, SA-64701-2A

Product Introduction

This product is a goat polyclonal secondary antibody conjugated to AcelFluor 647 (AF647), specifically recognizing the heavy chain of mouse IgG2a. Secondary antibodies are essential reagents in immunofluorescence (IF), immunohistochemistry (IH), flow cytometry (FC), ELISA (E), and other immunodetection techniques, providing specific detection of primary antibodies bound to target antigens. AF647 is a bright, photostable far-red fluorescent dye with excitation/emission maxima of approximately 650/665 nm, compatible with standard Cy5/APC filter sets and 633-640 nm laser-equipped instruments. The antibody is isolated from antisera by immunoaffinity chromatography using antigens coupled to agarose beads, ensuring high specificity and minimal cross-reactivity. By immunoelectrophoresis and ELISA, this antibody reacts specifically with mouse IgG2a heavy chain.

Product Features

Protocol (For Reference Only)

Important: Protect fluorescent conjugate from light. Centrifuge the vial briefly before opening. Optimal working dilution should be determined empirically for each application.

1. Immunofluorescence (IF): Fix and permeabilize samples according to standard protocols. Block with appropriate blocking buffer. Incubate with mouse IgG2a primary antibody. Wash and incubate with AF647-conjugated secondary antibody at 1:100 to 1:1,000 dilution for 1 hour at room temperature in the dark. Wash and mount with anti-fade mounting medium.

2. Immunohistochemistry (IH): Process tissue sections according to standard protocols. Following mouse IgG2a primary antibody incubation and washing, apply AF647-conjugated secondary antibody at 1:100 to 1:1,000 dilution for 30-60 minutes at room temperature in the dark. Wash and mount.

3. Flow Cytometry (FC): Following cell staining with mouse IgG2a primary antibody, wash cells and incubate with AF647-conjugated secondary antibody at 1:1,000 to 1:4,000 dilution for 30 minutes at 4¡ãC in the dark. Wash and analyze using a 633-640 nm laser with appropriate emission filter.

4. ELISA (E): Dilute the AF647-conjugated secondary antibody according to assay requirements and incubate following primary antibody binding. Detect according to the specific ELISA format.

Precautions

1. Protect from light to prevent photobleaching of the AF647 fluorophore.

2. Avoid freeze/thaw cycles; aliquot upon delivery and store at -20¡ãC.

3. The recommended dilutions are guidelines; optimal dilution must be determined empirically for each application.

4. Sodium azide is included as a preservative; handle with appropriate precautions.

5. For research use only. Not for use in diagnostic or therapeutic procedures.
