Dipeptidyl Peptidase IV (DPP4) Inhibitor Activity Fluorometric Assay Kit, IC-6577

Product Introduction

Dipeptidyl peptidase 4 (DPP4, EC 3.4.14.5), also known as CD26, is a type II transmembrane serine protease widely distributed on the surface of human cells. It cleaves peptide bonds at the second proline or alanine residue from the N-terminus of peptide chains. In vivo, DPP4 rapidly degrades incretins that promote blood glucose reduction. DPP4 inhibitors are a class of oral antidiabetic drugs that regulate blood glucose by inhibiting DPP4 activity. DPP4 cleaves its substrate to release the fluorescent compound AMC. This kit measures the change in the rate of fluorescence increase to calculate the inhibitory activity of DPP4 inhibitors.

Protocol (For Reference Only)

Important: This product is for research use only. Equilibrate reagents to the appropriate temperature before use. Prepare working solutions fresh according to the kit instructions.

1. Sample Preparation: Prepare animal tissue homogenates or serum/plasma samples according to standard protocols. Centrifuge to remove debris and collect the supernatant for analysis.

2. Reagent Preparation: Prepare the reaction working solution according to the kit instructions. Equilibrate reagents to the assay temperature before use.

3. Reaction Setup: Add samples, controls, and reaction working solution to the designated wells of a black 96-well plate according to the kit instructions.

4. Detection: Measure fluorescence at Ex 360 nm/Em 460 nm using a fluorescence microplate reader. Record fluorescence values at appropriate time intervals.

5. Calculation: Calculate the rate of fluorescence increase and determine DPP4 inhibitor activity according to the kit instructions.
