DPBS, without Calcium & Magnesium, without Phenol Red, IC-2014-S

Product Introduction

This product is a sterile-filtered Dulbecco''''s Phosphate-Buffered Saline (DPBS) formulated without calcium, magnesium, and phenol red. DPBS is a balanced salt solution widely used in cell culture and molecular biology for washing cells, diluting reagents, and as a base for cell suspension and transport. The absence of calcium and magnesium ions makes this formulation particularly suitable for cell dissociation and washing steps prior to trypsinization, as divalent cations can inhibit trypsin activity and promote cell clumping. The omission of phenol red eliminates potential estrogenic effects of the pH indicator and avoids interference with fluorescence-based assays and imaging applications. This DPBS is manufactured under rigorous quality control standards and is isotonic and non-toxic to mammalian cells, providing a reliable and versatile buffer system for routine laboratory procedures.

Product Features

1. Calcium and Magnesium-Free: Absence of divalent cations prevents cell clumping and enhances trypsin/EDTA effectiveness during cell dissociation procedures.

2. Phenol Red-Free: Eliminates potential estrogenic effects and avoids interference with fluorescence microscopy, flow cytometry, and colorimetric assays.

3. Sterile and Ready-to-Use: Supplied as a sterile-filtered (0.1 µm) solution requiring no additional preparation or dilution.

4. Endotoxin-Tested: Low endotoxin levels ensure compatibility with sensitive cell culture applications.

5. Versatile Application: Suitable for washing cells, diluting reagents, preparing cell suspensions, and serving as a base for transport media.

Specifications

Size: 500 mL

Formulation: Without Calcium Chloride, without Magnesium Chloride, without Phenol Red

pH: 7.2-7.4

Osmolality: 270-300 mOsm/kg

Storage and Stability

Storage Conditions: Store at room temperature (15-25¡ãC). Protect from light for long-term storage.

Shelf Life: The product is stable for 24 months from the date of manufacture when stored as directed in unopened packaging. Once opened, use within 4-6 weeks when stored properly and handled aseptically.

Protocol (For Reference Only)

Important: Use aseptic technique when handling to maintain sterility. Warm to room temperature or 37¡ãC before use for sensitive cell applications.

1. Cell Washing Prior to Trypsinization: Aspirate culture medium from adherent cells, add sufficient DPBS to cover the cell monolayer (typically 5-10 mL for a T75 flask), gently rock the flask to rinse the cells, aspirate the DPBS, and proceed with trypsin/EDTA treatment.

2. Cell Pellet Washing: Following centrifugation, aspirate the supernatant, resuspend the cell pellet in an appropriate volume of DPBS (typically 5-10 mL), gently pipette to achieve a single-cell suspension, centrifuge at 300-400 x g for 5 minutes, and discard the supernatant.

3. Reagent and Antibody Dilution: Use DPBS as a diluent for preparing working solutions of antibodies, dyes, or other reagents for cell staining and labeling applications.

4. Cell Suspension for Flow Cytometry: After trypsinization and neutralization, wash cells twice with DPBS, resuspend in DPBS at a concentration of 1¡Á10⁶-1¡Á10⁷ cells/mL, and filter through a 40-70 µm cell strainer before flow cytometric analysis.

5. Tissue Transport and Dissection: Use ice-cold DPBS for temporary storage and transport of freshly excised tissues and for dissection and processing of tissues for primary cell isolation.

Precautions

1. This formulation does not contain calcium, magnesium, or phenol red; verify formulation compatibility with experimental requirements before use.

2. Do not use DPBS as a long-term cell culture medium or incubation buffer as it lacks nutrients and buffering capacity for extended incubations.

3. Avoid prolonged exposure of cells to DPBS at room temperature or 37¡ãC as prolonged incubation may compromise cell viability.

4. If microbial contamination is suspected, discard the product and do not re-filter or re-sterilize.

5. For research use only. Not for use in diagnostic or therapeutic procedures.

FAQ (Simplified)

Q1: Why should I use DPBS without calcium and magnesium?

A1: Calcium and magnesium-free DPBS is recommended for washing cells prior to trypsinization because divalent cations inhibit trypsin activity and promote calcium-dependent cell adhesion, leading to incomplete dissociation and cell clumping.

Q2: Can I use this DPBS for long-term cell incubation?

A2: No. DPBS is a wash buffer and not a culture medium. It lacks nutrients, growth factors, and sufficient buffering capacity to maintain cell viability during extended incubations. Limit DPBS exposure to short-term washing and handling steps.

Q3: Is this DPBS compatible with fluorescence-based applications?

A3: Yes. The absence of phenol red eliminates autofluorescence and interference with fluorescence microscopy and flow cytometry, making this formulation ideal for fluorescence-based applications.

Q4: How is DPBS different from PBS?

A4: DPBS (Dulbecco''''s PBS) generally contains lower phosphate concentrations than standard PBS and may include potassium chloride and potassium phosphate monobasic, providing a more physiologically balanced ionic composition for mammalian cell applications.
