Recombinant Human Interleukin-18, CYT-Q14116
Product Introduction
Interleukin-18 (IL-18) is a secreted protein belonging to the IL-1 family. IL-18 is a proinflammatory cytokine produced by macrophages and other cells. This cytokine can induce IFN-¦Ã production by T cells. The combination of IL-18 and IL-12 has been shown to inhibit IL-4-dependent IgE and IgG1 production and enhance IgG2a production by B cells. IL-18 binding protein (IL18BP) can specifically interact with IL-18 and negatively regulate its biological activity. After stimulation with IL-18, natural killer (NK) cells and certain T cells release another important cytokine, interferon-¦Ã (IFN-¦Ã) or type II interferon, which plays an important role in activating macrophages and other cells.

Protocol (For Reference Only)
Important: Always centrifuge tubes before opening. Do not mix by vortex or pipetting. It is not recommended to reconstitute to a concentration less than 100 µg/mL.
1. Reconstitution: Dissolve the lyophilized protein in distilled water. Allow to dissolve completely with gentle mixing; do not vortex.
2. Aliquoting: Aliquot the reconstituted solution into single-use volumes to minimize freeze-thaw cycles.
3. Storage: Store aliquots at ¡Ü -20¡ãC for up to 3 months. Reconstituted solution may be stored at 2-8¡ãC for 2-7 days.
4. Activity Assay: Biological activity is measured by its ability to enhance IFN-¦Ã secretion in NK-92 human natural killer lymphoma cells, with an ED50 of 0.5-2.5 ng/mL.
5. Working Concentration: Optimal working concentrations should be determined empirically for each cell type and experimental system.
Precautions
1. Always centrifuge tubes before opening.
2. Do not mix by vortex or pipetting; gentle inversion or swirling is recommended.
3. It is not recommended to reconstitute to a concentration less than 100 µg/mL to avoid protein loss.
4. Aliquot reconstituted solution to minimize freeze-thaw cycles.
5. For research use only. Not for use in diagnostic or therapeutic procedures.
FAQ (Simplified)
Q1: How should I reconstitute this protein?
A1: Dissolve the lyophilized protein in distilled water to a concentration of at least 100 µg/mL. Do not vortex or pipette vigorously.
Q2: What is the biological activity of this protein?
A2: The ED50 is 0.5-2.5 ng/mL as measured by its ability to enhance IFN-¦Ã secretion in NK-92 human natural killer lymphoma cells.
Q3: How should I store the reconstituted protein?
A3: Reconstituted solution can be stored at 2-8¡ãC for 2-7 days. For longer storage, aliquot and store at ¡Ü -20¡ãC for up to 3 months. Avoid repeated freeze-thaw cycles.
Q4: What is the endotoxin level?
A4: <0.01 EU/µg as determined by LAL test.
