Recombinant Human IL-12, CYT-P29459
Product Introduction
IL-12 is a heterodimeric pleiotropic cytokine composed of a 40 kDa (p40) subunit and a 35 kDa (p35) subunit. Human and mouse IL-12 share 70% and 60% amino acid sequence identity in their p40 and p35 subunits, respectively. IL-12 is involved in the differentiation of naive T cells into Th1 cells. It is known as a T cell-stimulating factor that can stimulate the growth and function of T cells. It stimulates the production of interferon-gamma (IFN-¦Ã) and tumor necrosis factor-alpha (TNF-¦Á) from T cells and natural killer (NK) cells, and reduces IL-4-mediated suppression of IFN-¦Ã. T cells that produce IL-12 have a coreceptor, CD30, which is associated with IL-12 activity. IL-12 plays an important role in the activities of natural killer cells and T lymphocytes. IL-12 mediates enhancement of the cytotoxic activity of NK cells and CD8+ cytotoxic T lymphocytes.

Protocol (For Reference Only)
Important: Always centrifuge tubes before opening. Do not mix by vortex or pipetting. It is not recommended to reconstitute to a concentration less than 100 µg/mL.
1. Reconstitution: Dissolve the lyophilized protein in distilled water. Allow to dissolve completely with gentle mixing; do not vortex.
2. Aliquoting: Aliquot the reconstituted solution into single-use volumes to minimize freeze-thaw cycles.
3. Storage: Store aliquots at ¡Ü -20¡ãC for up to 3 months. Reconstituted solution may be stored at 2-8¡ãC for 2-7 days.
4. Activity Assay: Biological activity is measured by its ability to enhance IFN-¦Ã secretion in NK-92 human natural killer lymphoma cells, with an ED50 of 0.2-0.8 ng/mL.
5. Working Concentration: Optimal working concentrations should be determined empirically for each cell type and experimental system.
Precautions
1. Always centrifuge tubes before opening.
2. Do not mix by vortex or pipetting; gentle inversion or swirling is recommended.
3. It is not recommended to reconstitute to a concentration less than 100 µg/mL to avoid protein loss.
4. Aliquot reconstituted solution to minimize freeze-thaw cycles.
5. For research use only. Not for use in diagnostic or therapeutic procedures.
FAQ (Simplified)
Q1: How should I reconstitute this protein?
A1: Dissolve the lyophilized protein in distilled water to a concentration of at least 100 µg/mL. Do not vortex or pipette vigorously.
Q2: What is the biological activity of this protein?
A2: The ED50 is 0.2-0.8 ng/mL as measured by its ability to enhance IFN-¦Ã secretion in NK-92 human natural killer lymphoma cells.
Q3: How should I store the reconstituted protein?
A3: Reconstituted solution can be stored at 2-8¡ãC for 2-7 days. For longer storage, aliquot and store at ¡Ü -20¡ãC for up to 3 months. Avoid repeated freeze-thaw cycles.
Q4: What is the subunit composition of this protein?
A4: IL-12 is a heterodimeric cytokine composed of a 40 kDa (p40) subunit and a 35 kDa (p35) subunit.
