Sodium Citrate Antigen Retrieval Solution

pH 6.0, 50X, Mild Formulation, IC-20010

Product Introduction

This product is a 50X concentrated sodium citrate antigen retrieval solution (pH 6.0) with a mild formulation, designed for heat-induced epitope retrieval (HIER) in immunohistochemistry (IHC) and immunocytochemistry (ICC) applications. HIER is used to reverse the loss of immunoreactivity that occurs in some epitopes during formalin fixation and paraffin embedding of tissues. During antigen retrieval, tissue slides are immersed in the solution and heated using a hot plate (>95¡ãC) or other suitable heat sources, such as a pressure cooker or steamer. Citrate buffer is the preferred solution for the majority of antibodies. This product is supplied as a 50X concentrate and should be diluted to 1X working solution with distilled water before use.

Product Features

1. Standard HIER Buffer: Widely used sodium citrate antigen retrieval solution at pH 6.0 for IHC and ICC applications.

2. Mild Formulation: Gentle retrieval conditions help preserve tissue morphology and reduce the risk of section detachment.

3. 50X Concentrated Format: Convenient storage and cost-effective; dilute 50-fold before use.

4. Broad Antibody Compatibility: Citrate buffer is the preferred retrieval solution for most antibodies.

5. Ready-to-Dilute: Simple dilution with distilled water for 1X working solution preparation.

Specifications

Size: 100 mL / 500 mL

Concentration: 50X

pH: 6.0

1X Working Solution: Sodium citrate buffer, pH 6.0

Application: Heat-induced epitope retrieval for IHC and ICC

Storage and Stability

Storage Conditions: Store at room temperature (15-25¡ãC). If precipitation occurs at low temperatures, warm to room temperature and mix before use.

Shelf Life: The product is stable for 24 months from the date of manufacture when stored as directed.

Protocol (For Reference Only)

Important: Ensure any precipitate is fully dissolved before dilution. Prepare 1X working solution fresh or store at 4¡ãC for short-term use.

1. Preparation of 1X Working Solution: Dilute the 50X Sodium Citrate Antigen Retrieval Solution 1:50 with distilled water (e.g., 10 mL of 50X concentrate plus 490 mL of distilled water to prepare 500 mL of 1X working solution). Mix thoroughly. Adjust pH to 6.0 if necessary.

2. Deparaffinization and Rehydration: Deparaffinize tissue sections in xylene and rehydrate through graded ethanol to distilled water according to standard protocols.

3. Heat-Induced Antigen Retrieval:

- Hot plate method: Place slides in a container filled with 1X antigen retrieval solution and heat on a hot plate (>95¡ãC) for 20-40 minutes. Allow slides to cool in the solution at room temperature for 20-30 minutes.

- Pressure cooker method: Heat slides in 1X antigen retrieval solution in a pressure cooker for 2-5 minutes after reaching maximum pressure. Allow to cool and release pressure before opening.

- Steamer method: Place slides in a container filled with 1X antigen retrieval solution and steam for 20-40 minutes. Allow to cool at room temperature for 20-30 minutes.

4. Washing: Wash slides 3 times with PBS or TBS for 5 minutes each.

5. Subsequent Staining: Proceed with blocking, primary antibody incubation, and standard IHC detection procedures.

Precautions

1. If precipitation occurs during storage, warm to room temperature and mix until fully dissolved.

2. Do not use the 50X concentrate directly; always dilute to 1X before use.

3. Heating time and method should be optimized based on tissue type and antigen. Over-retrieval may cause tissue damage or false-positive staining; under-retrieval may result in weak or absent signal.

4. Allow slides to cool gradually in the retrieval solution to prevent tissue detachment.

5. For research use only. Not for use in diagnostic or therapeutic procedures.

FAQ (Simplified)

Q1: How do I prepare 1X working solution from the 50X concentrate?

A1: Dilute the 50X concentrate 1:50 with distilled water (e.g., 10 mL of 50X concentrate plus 490 mL of distilled water for 500 mL of 1X working solution).

Q2: What is the purpose of antigen retrieval?

A2: Antigen retrieval unmasks epitopes that are masked by formaldehyde fixation-induced cross-links, restoring antibody accessibility and enabling sensitive detection of target proteins.

Q3: What heating methods can be used?

A3: Hot plate (>95¡ãC), pressure cooker, or steamer may be used. Optimize heating time and method for each tissue type and antigen.

Q4: How should I store this product?

A4: Store at room temperature. If precipitation occurs, warm to room temperature and mix before use.

Disclaimer

1. For Research Use Only. Not for use in diagnostic or therapeutic procedures.

2. Due to the variable nature of biological research, optimization of retrieval conditions is recommended for specific tissue types and antigens.
