Mouse Peripheral Blood Mononuclear Cell
Separation Medium Kit, IC-1192
Product Introduction
This product is a sterile, low-endotoxin density gradient separation medium designed for the isolation of animal peripheral blood mononuclear cells (PBMCs). The separation principle is based on differences in the density of blood cells. Through centrifugation, cells of a specific density distribute along the corresponding density gradient, allowing mononuclear cells to be separated from peripheral blood. It is suitable for isolating mononuclear cells from anticoagulated animal blood, and mononuclear cells isolated under aseptic conditions can be used for culture.
Product Features
1. Density Gradient Separation: Separates mononuclear cells from whole blood based on density differences of blood cell populations.
2. Sterile and Low Endotoxin: Suitable for downstream cell culture applications.
3. Optimized for Mouse Blood: Formulated for the isolation of mouse peripheral blood mononuclear cells.
4. Ready-to-Use: Supplied as a sterile solution requiring no additional preparation.
Specifications
Size: 100 mL per kit / 200 mL per kit
Application: Isolation of mouse peripheral blood mononuclear cells
Form: Sterile liquid separation medium
Storage and Stability
Storage Conditions: Store at 4-8¡ãC, protected from light. Do not freeze. Equilibrate to room temperature before use.
Shelf Life: The product is stable for 24 months from the date of manufacture when stored as directed.
Protocol (For Reference Only)
Important: Use aseptic technique throughout the procedure. Allow the separation medium to equilibrate to room temperature (18-25¡ãC) before use. All centrifugation steps should be performed at room temperature with the brake off to preserve the gradient interface.
1. Blood Sample Preparation: Collect mouse peripheral blood in anticoagulant tubes (EDTA or heparin). Dilute the blood with an equal volume of sterile PBS or RPMI-1640 medium without serum and mix gently.
2. Gradient Setup: Add an appropriate volume of separation medium to a centrifuge tube. Carefully layer the diluted blood over the separation medium without disturbing the interface.
3. Density Gradient Centrifugation: Centrifuge at 400-500 x g for 20-30 minutes at room temperature with the brake off. After centrifugation, mononuclear cells will be visible as a thin white band at the interface between the plasma and separation medium layers.
4. PBMC Collection: Using a sterile pipette, carefully aspirate the mononuclear cell layer from the interface and transfer to a clean centrifuge tube. Avoid aspirating the separation medium to minimize granulocyte contamination.
5. Cell Washing: Add 3-5 volumes of sterile PBS or RPMI-1640 to the collected cells, mix gently, and centrifuge at 300 x g for 10 minutes at room temperature. Discard the supernatant, resuspend the cell pellet in fresh medium or buffer, and repeat the wash step once or twice to remove residual separation medium and platelets.
6. Downstream Applications: Resuspend the final cell pellet in appropriate medium or buffer for downstream applications including cell culture, flow cytometry, and functional assays.
Precautions
1. Allow the separation medium to reach room temperature before use; cold medium has altered density that reduces separation efficiency.
2. Layer the diluted blood carefully over the separation medium without disrupting the interface.
3. Centrifugation with the brake off is essential to preserve the mononuclear cell band at the interface.
4. Minimize platelet contamination by washing cells at least twice.
5. For research use only. Not for use in diagnostic or therapeutic procedures.
FAQ (Simplified)
Q1: What is the principle of mononuclear cell separation?
A1: The separation medium creates a density gradient that allows mononuclear cells to be separated from other blood cell populations based on differences in cell density during centrifugation.
Q2: Can the isolated cells be used for culture?
A2: Yes. Mononuclear cells isolated under aseptic conditions can be used for cell culture and other downstream applications.
Q3: Why must the centrifuge brake be turned off?
A3: The brake causes rapid deceleration that generates turbulence, disrupting the delicate mononuclear cell band at the density interface.
Q4: How should I store this product?
A4: Store at 4-8¡ãC protected from light. Do not freeze. Equilibrate to room temperature before use.
