0.1% Crystal Violet Staining Solution, IC-21001
Product Introduction
This product is a 0.1% crystal violet staining solution, widely used for the staining of cell nuclei, cell colonies, and tissue sections in biological research. Crystal violet is a basic triphenylmethane dye that binds to acidic components of cells, particularly DNA, producing a distinctive purple color. In cell biology, 0.1% crystal violet staining solution is commonly used for colony formation assays to visualize and quantify adherent cell colonies, for cell morphology observation, and for the assessment of cell migration and invasion in Transwell assays. The ready-to-use formulation provides convenient and reproducible staining for routine laboratory applications.

Protocol (For Reference Only)
Important: Use appropriate personal protective equipment when handling crystal violet solution, as it may stain skin and clothing.
1. Colony Formation Assay: After colony formation, remove culture medium and wash cells gently with PBS. Fix cells with 4% paraformaldehyde or methanol for 10-15 minutes. Remove fixative and add sufficient 0.1% crystal violet staining solution to cover the cells. Stain for 10-20 minutes at room temperature. Remove staining solution and wash gently with PBS or distilled water until background is clear. Air dry and photograph or count colonies.
2. Transwell Migration/Invasion Assay: After the assay, remove non-migrated cells from the upper surface of the membrane with a cotton swab. Fix migrated cells with 4% paraformaldehyde or methanol for 10-15 minutes. Stain with 0.1% crystal violet staining solution for 10-20 minutes at room temperature. Wash and air dry. Quantify migrated cells by microscopy or elute the dye with 10% acetic acid and measure absorbance at 560-590 nm.
3. Cell Morphology Observation: Wash cells with PBS, fix with 4% paraformaldehyde or methanol, and stain with 0.1% crystal violet staining solution for 10-20 minutes. Wash and observe under a microscope.
Precautions
1. Crystal violet may stain skin and clothing; wear gloves and laboratory protective clothing when handling.
2. Protect from light during storage.
3. The staining solution may be reused several times; filter if particulate matter accumulates.
4. Dispose of crystal violet waste according to institutional guidelines.
5. For research use only. Not for use in diagnostic or therapeutic procedures.
