Description 

ExperPro WesternBlot Fast Blocking Buffer contains a proprietary non-relevant protein for blocking excess binding sites in  Western blotting applications. This blocking buffer has been optimized for use with precipitating substrates and yields a high signal-to-noise ratio in most applications. Additionally, this blocking buffer does not contain albumin or endogenous biotin and, therefore, may be used with avidin/streptavidin systems.Because no blocking reagent is optimal for all systems, empirical testing is essential to determine the appropriate blocking buffer for each Western blot system. Determining the proper blocking buffer can increase sensitivity and prevent nonspecific signal caused by cross-reactivity between the antibody and the blocking reagent.

This product has added preservatives that do not affect the activity of HRP and AP, and will not interfere with the detection of HRP or AP labeled secondary antibodies. At the same time, this product does not contain biotin and will not interfere with biotin based detection.

Usage

1. Remove blot from the transfer unit.

2. Use a sufficient volume of blocking buffer to completely cover blot. Using large volumes of blocking and wash buffer often reduces nonspecific signal.

3. Incubate for approximately 5-15 minutes with agitation.

4.Remove the blocking buffer and continue Western blot procedure.

Storage

The product will be stable for 12 months under 2-8¡æ.

Troubleshooting


Problem

No signal or weak signal

Poor transfer efficiency

Optimize the membrane transferring procedure

Insufficient antigen

Increase the amount of loaded antigen

Make sure the blot have been store correctly to avoid the degradation of target protein

The concentration of primary and secondary antibody is too low

Increase the concentration of the primary and/or the secondary antibody

Inappropriate storage/preparation of the ECL detection reagents

Use HRP or HRP conjugates to check the applicability of ECL reagents

Too short exposure time

Optimize the condition by reducing the amount of antigen, or the concentration of the primary antibody and/or secondary antibody. Initially, reduce the secondary antibody to 20% of the original usage
