 BSA£¨Cell Culture Grade£©, IC-1305

Product Introduction

This product is a high-purity bovine serum albumin (BSA) specifically processed to achieve exceptionally low endotoxin levels, making it suitable for sensitive cell culture applications, in vivo studies, and immunological assays where endotoxin contamination must be rigorously excluded. BSA is a 66 kDa serum protein that functions as a carrier for hydrophobic molecules, a nutrient source in cell culture, a blocking agent in immunoassays, and a stabilizer for enzymes and antibodies. Standard BSA preparations may contain significant levels of endotoxins (lipopolysaccharides) that activate toll-like receptor 4 (TLR4) signaling, inducing pro-inflammatory cytokine production and confounding results in cell-based and in vivo experiments. This low endotoxin BSA is manufactured under strictly controlled conditions and tested to ensure endotoxin levels below 0.1 EU/mg, providing a reliable protein source for endotoxin-sensitive applications including primary cell culture, stem cell research, macrophage and monocyte studies, and in vivo animal experiments.

Product Features

1. Low Endotoxin Levels: Certified endotoxin content ¡Ü0.1 EU/mg, minimizing TLR4-mediated inflammatory responses in sensitive cell and in vivo systems.

2. High Purity: Supplied at ¡Ý98% purity as determined by SDS-PAGE, with minimal contaminating immunoglobulins and other serum proteins.

3. Versatile Applications: Suitable as a cell culture supplement, blocking agent in ELISA and Western blot, carrier protein for dilute enzyme and antibody solutions, and stabilizer in molecular biology reactions.

4. Fatty Acid-Free Option Characteristics: Processed to reduce fatty acid content, enhancing suitability for lipid metabolism and hormone studies.

5. Rigorous Quality Control: Each lot is tested for endotoxin levels, purity, pH, solubility, and performance in cell culture to ensure batch-to-batch consistency.

Protocol (For Reference Only)

Important: Prepare BSA solutions freshly and filter sterilize for cell culture applications. Avoid repeated freeze-thaw cycles of reconstituted solutions.

1. Stock Solution Preparation for Cell Culture: Dissolve low endotoxin BSA in sterile PBS or culture medium to the desired concentration (typically 1-10% w/v). Filter sterilize through a 0.22 µm membrane before addition to cell culture medium.

2. Blocking Buffer for ELISA and Western Blot: Prepare 1-5% (w/v) BSA in PBS or TBS with 0.05-0.1% Tween-20. For low background applications, use 1-3% BSA. Filter through a 0.45 µm membrane if turbidity is observed.

3. Carrier Protein for Dilute Enzyme and Antibody Solutions: Add BSA to a final concentration of 0.1-1% (w/v) to stabilize dilute protein solutions and prevent non-specific adsorption to container surfaces.

4. Cell Culture Supplement: Supplement serum-free or low-serum culture medium with 0.1-1% (w/v) low endotoxin BSA as a defined protein source. For stem cell and primary cell applications, titrate the optimal concentration empirically.

5. In Vivo Applications: For in vivo administration, dissolve low endotoxin BSA in sterile physiological saline or PBS at the desired concentration. Filter sterilize and use promptly. Typical concentrations for in vivo carrier or vehicle applications range from 0.1-5% (w/v).

Precautions

1. Use aseptic technique when preparing BSA solutions for cell culture or in vivo use to maintain low endotoxin status.

2. Filter sterilize solutions through a 0.22 µm membrane; do not autoclave as this may denature the protein and increase endotoxin bioavailability.

3. Store reconstituted BSA solutions at 4¡ãC and use within 1-2 weeks; for longer storage, aliquot and freeze at -20¡ãC.

4. This product is not sterile; if required for sterile applications, filter sterilize the prepared solution.

5. For research use only. Not for use in diagnostic or therapeutic procedures.

FAQ (Simplified)

Q1: What endotoxin level is considered "low endotoxin"?

A1: This product is certified to contain ¡Ü0.1 EU/mg of endotoxin, which is significantly lower than standard BSA preparations that may contain 1-10 EU/mg or higher. This level is suitable for most cell culture and in vivo applications where endotoxin-induced TLR4 activation must be avoided.

Q2: How does low endotoxin BSA differ from standard BSA?

A2: Low endotoxin BSA is processed under controlled conditions to minimize endotoxin contamination. Standard BSA preparations may contain endotoxin levels sufficient to activate TLR4 signaling in sensitive cells such as macrophages, monocytes, dendritic cells, and primary cells, potentially confounding experimental results.

Q3: Can I use this product for cell culture?

A3: Yes. This low endotoxin BSA is particularly suitable for cell culture applications, especially for primary cells, stem cells, and immune cells that are sensitive to endotoxin-mediated activation. Prepare solutions aseptically and filter sterilize before use.

Q4: Is this BSA fatty acid-free?

A4: This BSA is processed to have reduced fatty acid content characteristic of purified BSA preparations. For applications specifically requiring certified fatty acid-free BSA, please inquire separately or verify with the lot-specific certificate of analysis.
